Galectin-3 is a glycan-binding protein identified as a key regulator of neuroinflammation in Alzheimer’s disease. The research potential for AD is clear:

  • AD patients have significantly increased galectin-3 levels

  • Mice without galectin-3 show less amyloid plaque deposition and better cognitive function

  • No potent galectin-3 inhibitors currently pass the blood-brain barrier

GALE will infuse a highly specific galectin-3 inhibitor directly into the brains of AD mice over four weeks, monitoring effects on neuroinflammation and plaque deposition.

If successful, this validates galectin-3 as a target for pharmacological intervention. It could yield results not just for AD but for an array of neurodegenerative diseases.

The team

The project is led by key contributors of the Cerebrum DAO team:

  • Dr. Maryna Polyakova: Neuroscientist and lecturer at HTW Berlin, University of Leipzig, and Max-Planck Research School. Uses neuroimaging, neuropsychological testing, and machine learning to improve early diagnostics. Leads the Neurotech Tree project at Foresight Institute, building a new roadmap for the future of neurotechnology.

  • Dr. Peter Groenen: Industry executive with deep expertise in pharmaceutical R&D, specializing in translational science and biomarker integration. Experienced in driving projects from early research to late-stage development, collaborating with academia, industry, and regulatory bodies to accelerate therapeutic innovation.

The opportunity

The brain health market is valued in the trillions annually, with 150 million AD cases expected globally by 2050. Current treatments are minimally effective at best.

Galectin-3 is a novel target in AD. A disease-modifying inhibitor with a compelling safety profile could attain blockbuster drug status, especially for early-stage or pre-symptomatic populations.

Roadmap

Q1 2026

  • In vivo proof-of-concept in mouse AD model using Alzet pump with galectin-3 inhibitor

  • Brain analysis via ELISA/Western Blot measuring inflammation markers and Aß42 levels

Q2 2026

  • Immunohistochemistry analysis: plaque load and microglia analysis (Iba1, Gal3)

Q3 2026

  • Complete study data analysis combining all brain analysis data to identify clear inhibitory effects on AD descriptors
